Summary
Sample preparation
Parsley seed was dried and milled before extraction; the raw-material charge was weighed and its moisture content determined so the run could be reported on a dry basis.
Extraction
Extraction used pure supercritical CO2 with no co-solvent on the SuperEx SC-1000 lab-scale system in dynamic mode, in a two-stage isothermal run at 40 °C. Stage 1 ran at 100 bar and stage 2 at 400 bar. The two stage extracts were characterised separately and then pooled and reported as the total extract, so the contribution of each pressure stage stays visible.
Analysis
Each stage was run by qualitative GC-MS and quantitative GC-FID, with extract aliquots of roughly 10 mg in 1 mL n-hexane injected directly. Fatty acid methyl esters were quantified against an external five-point calibration. Total phenolic content was determined as gallic-acid equivalents, and antioxidant activity as DPPH radical scavenging with IC50 obtained by linear interpolation.
